چکیده

A continuous wave and pulse electron paramagnetic resonance study of the base-on form of cob(II)alamin diluted in hydroxocob(III)alamin powder is presented. HYSCORE spectroscopy at Xand Q-band was used to study the weakly coupled corrin nitrogens and the remote nitrogen of the axial dimethylbenzimidazole ligand. Simulations of the spectra measured at different field positions enabled the following parameters to be determined; for the corrin nitrogens the hyperfine principal values are A1 1⁄4 4:5 MHz, A2 1⁄4 3:4 MHz and A3 1⁄4 ð 2:2 to )2.5) MHz with Euler angles 1⁄2a; b; c 1⁄4 1⁄2n90; 25; 0 and the nuclear quadrupole parameters are eqQ=h 1⁄4 1:75 MHz and g 1⁄4 0:95 with Euler angles 1⁄2a; b; c 1⁄4 1⁄290þ n90; 90; 12:5 , where n 1⁄4 0; 1; 2; 3. The remote nitrogen of the dimethylbenzimidazole ligand has A1 1⁄4 1:75 MHz, A2 1⁄4 1:75 MHz and A3 1⁄4 2:10 MHzwith Euler angles 1⁄2a; b; c 1⁄4 1⁄2 45; 10; 0 and eqQ=h 1⁄4 3:3 MHz and g 1⁄4 0:1 with Euler angles 1⁄2a; b; c 1⁄4 1⁄245; 90; 67 . 2002 Elsevier Science B.V. Residue Phe112 of the human-type corrinoid adenosyltransferase (PduO) enzyme of Lactobacillus reuteri is critical to the formation of the four-coordinate Co(II) corrinoid substrate and to the activity of the enzyme.

ATP:Corrinoid adenosyltransferases (ACAs) catalyze the transfer of the adenosyl moiety from ATP to cob(I)alamin via a four-coordinate cob(II)alamin intermediate. At present, it is unknown how ACAs promote the formation of the four-coordinate corrinoid species needed for activity. The published high-resolution crystal structure of the ACA from Lactobacillus reuteri (LrPduO) in complex with ATP a...

Structural insights into the mechanism of four-coordinate Cob(II)alamin formation in the active site of the Salmonella enterica ATP:Co(I)rrinoid adenosyltransferase enzyme: critical role of residues Phe91 and Trp93.

ATP:co(I)rrinoid adenosyltransferases (ACATs) are enzymes that catalyze the formation of adenosylcobalamin (AdoCbl, coenzyme B(12)) from cobalamin and ATP. There are three families of ACATs, namely, CobA, EutT, and PduO. In Salmonella enterica, CobA is the housekeeping enzyme that is required for de novo AdoCbl synthesis and for salvaging incomplete precursors and cobalamin from the environment...

Characterisation of PduS, the pdu Metabolosome Corrin Reductase, and Evidence of Substructural Organisation within the Bacterial Microcompartment

PduS is a corrin reductase and is required for the reactivation of the cobalamin-dependent diol dehydratase. It is one component encoded within the large propanediol utilisation (pdu) operon, which is responsible for the catabolism of 1,2-propanediol within a self-assembled proteinaceous bacterial microcompartment. The enzyme is responsible for the reactivation of the cobalamin coenzyme require...

Protein-coenzyme interactions in adenosylcobalamin-dependent glutamate mutase.

Glutamate mutase catalyses an unusual isomerization involving free-radical intermediates that are generated by homolysis of the cobalt-carbon bond of the coenzyme adenosylcobalamin (coenzyme B(12)). A variety of techniques have been used to examine the interaction between the protein and adenosylcobalamin, and between the protein and the products of coenzyme homolysis, cob(II)alamin and 5'-deox...

Kinetic studies on the reaction of cob(II)alamin with hypochlorous acid: Evidence for one electron oxidation of the metal center and corrin ring destruction.

Kinetic and mechanistic studies on the reaction of a major intracellular vitamin B12 form, cob(II)alamin (Cbl(II)), with hypochlorous acid/hypochlorite (HOCl/OCl-) have been carried out. Cbl(II) (Co(II)) is rapidly oxidized by HOCl to predominately aquacobalamin/hydroxycobalamin (Cbl(III), Co(III)) with a second-order rate constant of 2.4×107M-1s-1 (25.0°C). The stoichiometry of the reaction is...
